Tapentadol API (Nucynta / Palexia)

Semi-synthetic opioid API with dual mechanism: mu-opioid receptor agonism + norepinephrine reuptake inhibition (NRI). Developed by Grünenthal; licensed to Janssen Pharmaceuticals (J&J) as Nucynta in the US and marketed as Palexia in EU/ROW. Tapentadol offers comparable analgesia to oxycodone with reportedly lower constipation and lower abuse potential. Schedule II in the US.
