What Amicus Therapeutics, Inc. told the SEC could break it.
Amicus's sharpest exposure is on the supply side: it makes none of its products itself and relies on third-party manufacturers, and the majority of its candidates and commercial products — including both of its marketed rare-disease drugs, Galafold and Pombiliti — come from single-source CMOs with no qualified backup, so a stoppage at any of them would mean supply and development delays. Its sales are also concentrated, with one distributor or specialty pharmacy accounting for 24% of net product sales in 2025 and two customers each holding 12% of product-sales receivables. And because the majority of its Galafold and Pombiliti+Opfolda revenue comes from outside the U.S., its results are exposed to foreign-currency swings, exchange controls and varying ex-U.S. reimbursement regimes.
3 self-disclosed vulnerabilities, pulled from its own filings, each in the company’s words, with the source. This is the risk register almost nobody reads.
In its own words
What could break it.
Sole-source dependency
- Single-source third-party manufacturers for Galafold & Pombilitihigh
Amicus relies on third-party manufacturers for all of its products, and the majority of its product candidates and commercial products — including its two marketed rare-disease drugs Galafold and Pombiliti — are made by single-source CMOs; if any of those manufacturers stopped, Amicus would face supply and development delays with no qualified backup.
“The majority of our product candidates and commercial products, including Galafold ® and Pombiliti ® , are manufactured by single source third party manufacturers.”

Customer concentration
- One customer = 24% of net product sales (unnamed distributor/pharmacy)medium
Amicus has meaningful customer concentration: one customer (an unnamed distributor/specialty pharmacy) accounted for 24% of net product sales in 2025 (26% in 2024), and two customers each held 12% of product-sales receivables — so loss of that lead distributor relationship would disrupt a large share of revenue and collections.
“In 2025 and 2024, one customer accounted for 24 % and 26 % of net product sales, respectively.”

Geographic concentration
- Majority of Galafold & Pombiliti revenue from outside the U.S. (FX exposure)medium
The majority of Amicus' Galafold and Pombiliti+Opfolda revenue comes from outside the United States, exposing results to foreign-currency fluctuations, exchange controls and currency devaluations, as well as varying ex-US reimbursement regimes.
“The majority of our current Galafold ® and Pombiliti ® + Opfolda ® revenue is derived from outside the U.S.”
