Agenus (formerly Antigenics Inc.) was founded in 1994 by Pramod Srivastava, an immunologist who had developed heat shock protein (HSP)-based cancer vaccine concepts at Yale. The company initially focused on personalized cancer vaccines (Oncophage) but diversified by acquiring the rights to QS-21, a saponin adjuvant discovered by Charlotte Kensil at Cambridge Biotech in the early 1990s. Agenus developed the pharmaceutical-grade manufacturing specifications and characterization methods for QS-21 that made it suitable for human vaccine use, and then licensed it exclusively to GSK. The QS-21 licensing became Agenus's primary revenue source as Oncophage development struggled. When Shingrix launched in 2017 and became the most commercially successful vaccine in history at its launch, Agenus began collecting royalties on each Shingrix dose sold -- making a Lexington MA biopharma company with a struggling cancer vaccine pipeline a significant beneficiary of global shingles vaccination campaigns without being the vaccine developer.
