    Novavax's COVID-19 vaccine efficacy depends on Matrix-M, a proprietary adjuvant derived from the bark of the soapbark tree (Quillaja saponaria) — a tree native to the dry coastal slopes of Chile. The active ingredient is a purified saponin fraction called QS-21 (quillaja saponin-21), which stimulates both antibody and T-cell responses far more powerfully than aluminum-based adjuvants. The global supply of QS-21 saponins is extracted almost entirely from wild or semi-cultivated Quillaja saponaria forests in Chile, creating a South American agricultural supply chain as a hidden dependency for at least three Western COVID and shingles vaccines (Novavax Matrix-M, GSK AS01B in Shingrix, and GSK AS01E in Mosquirix all use QS-21-family saponins). Novavax sources Quillaja extract through Desert King International (a Chilean company), processes it into the ISCOM matrix structure that forms the Matrix-M particles. Climate stress, drought, and land-use change in the Chilean coastal cordillera represent an unmodeled climate-supply chain risk to global vaccine adjuvant supply that no manufacturer publicly discloses.

    Novavax's protein subunit vaccine platform uses a baculovirus expression system — the same insect-cell manufacturing technology used to produce flu vaccines (Flublok) and some oncology biologics. Sf9 moth cells (Spodoptera frugiperda, the fall armyworm) infected with recombinant baculovirus express the spike protein antigen at scale. This means Novavax's vaccine manufacturing capacity is fungible with flu vaccine production: the same bioreactor lines, the same insect-cell culture expertise, the same downstream purification steps apply to both. During COVID, Novavax's manufacturing ramp-up directly competed with Sanofi's Flublok flu vaccine production for the same contract manufacturing partners and Sf9 cell supply chains — a zero-sum trade-off between pandemic response and routine flu vaccination that public health planners did not publicly acknowledge.
