The process

  1. 01

    Solid-phase peptide synthesis (SPPS)

    The peptide backbone (e.g. semaglutide, a 31-residue analog) is assembled residue-by-residue on resin in large SPPS reactors.

  2. 02

    Side-chain acylation & resin cleavage

    The lysine side chain is acylated with a C18 fatty-diacid linker (via Ado spacers + γ-Glu), the unnatural Aib residue is incorporated, then the peptide is cleaved from resin.

  3. 03

    Chromatographic purification & lyophilization

    Crude peptide is purified by preparative HPLC and freeze-dried to drug-substance (API).

  4. 04

    Aseptic formulation & fill-finish

    API is formulated and aseptically filled into ready-to-use Type I borosilicate glass cartridges — the sterile fill-finish step.

  5. 05

    Pen / auto-injector device assembly

    Filled cartridges are assembled into disposable injection pens / autoinjectors, then labeled.

  6. 06

    Cold-chain packaging & distribution

    Finished pens are packaged and shipped under refrigerated cold-chain to pharmacies and patients.
