  1. 01

    Inoculum Preparation

    Penicillium chrysogenum or Streptomyces strains grown in seed media under controlled conditions to prepare viable inoculum for main fermentation.

  2. 02

    Submerged Fermentation

    Batch or fed-batch fermentation in large bioreactors (often 200,000+ liters in China). Media contains glucose, corn steep liquor, and side-chain precursors (phenylacetic acid for penicillin G, phenoxyacetic acid for penicillin V). Runs 5–7 days at 25–26°C.

  3. 03

    Harvest & Broth Filtration

    Centrifugation and filtration to separate fungal/bacterial biomass from fermentation broth containing the antibiotic.

  4. 04

    Solvent Extraction & Purification

    Acidification of broth to pH 2.0–2.5 and extraction with organic solvents (butyl acetate, amyl acetate). Charcoal treatment removes impurities; back-extraction into aqueous phase with NaOH.

  5. 05

    Crystallization & Drying

    Cooling or addition of precipitation agents produces solid antibiotic crystals (e.g., penicillin G potassium/sodium salt). Crystals washed and dried to pharmaceutical grade.

  6. 06

    Semi-Synthesis of API (for amoxicillin, ampicillin, etc.)

    Penicillin G enzymatically cleaved by Penicillin G Acylase (PGA) to produce 6-APA nucleus. 6-APA is then coupled with appropriate side-chain precursors to produce semi-synthetic penicillins (amoxicillin, ampicillin, piperacillin). Cephalosporins follow analogous 7-ACA route.

    Formulation

    API blended with excipients and formulated into tablets, capsules, oral suspension, or sterile injectable solution. Sterile products require aseptic fill-finish in classified cleanrooms.

  8. 08

    Quality Testing & Packaging

    Finished product tested against USP/BP monographs for identity, potency, sterility, and endotoxins. Packaged and labeled per regional regulatory requirements.
